Paclitaxel - TAXOL® und Generika

According to the NCI website paclitaxel is a compound extracted from the Pacific yew tree Taxus brevifolia with antineoplastic activity. Paclitaxel binds to tubulin and inhibits the disassembly of microtubules, thereby resulting in the inhibition of cell division. This agent also induces apoptosis by binding to and blocking the function of the apoptosis inhibitor protein Bcl-2 (B-cell Leukemia 2)..

Indikationen/Anwendungsmöglichkeiten gemäss Compendium®:

Ovarialkarzinom

  • Erstbehandlung von Ovarialkarzinomen in fortgeschrittenem Stadium (FIGO-Stadium III, IV) oder von Residualkarzinomen (>1 cm) nach Laparotomie, in Kombination mit Cisplatin.
  • Behandlung von metastasierenden Ovarialkarzinomen, wenn eine Standardtherapie mit platinhaltigen Chemotherapie-Kombinationen versagt hat.

Nicht-kleinzelliges Bronchialkarzinom

  • Behandlung von nicht-kleinzelligen Bronchialkarzinomen (NSCLC) in Kombination mit Cisplatin, wenn ein chirurgischer Eingriff und/oder eine Strahlenbehandlung nicht angezeigt sind.

Mammakarzinom

  • In Kombination mit Trastuzumab zur Behandlung von metastasierenden Mammakarzinomen, wenn die Tumoren HER 2 überexprimieren und die Patienten noch keine Chemotherapie gegen ihre metastasierende Erkrankung erhalten haben.
  • Behandlung von metastasierenden Mammakarzinomen, wenn eine anthrazyklinhaltige Standardtherapie versagt hat oder nicht indiziert ist.

Adjuvante Therapie bei Patientinnen mit Mammakarzinom und befallenen lokoregionalen Lymphknoten (nodal positiv) nach einer anthrazyklinhaltigen Behandlung mit Doxorubicin und Cyclophosphamid.

Taxane

Paclitaxel an Nanopartikel-Albumin gebunden - ABRAXANE®

According to the NCI website, abraxane is a paclitaxel albumin-stabilized nanoparticle formulation, a Cremophor EL-free, albumin-stabilized nanoparticle formulation of the natural taxane paclitaxel with antineoplastic activity. Paclitaxel binds to and stabilizes microtubules, preventing their depolymerization and so inhibiting cellular motility, mitosis, and replication. This formulation solubilizes paclitaxel without the use of the solvent Cremophor, thereby permitting the administration of larger doses of paclitaxel while avoiding the toxic effects associated with Cremophor. Indikationengemäss Compendium®
  • Abraxane ist indiziert für die Behandlung des metastasierenden Mammakarzinoms nach Versagen einer Chemotherapie oder Rezidiv innert 6 Monaten nach adjuvanter Chemotherapie. In der Vortherapie sollte ein Anthrazyklin enthalten gewesen sein, es sei denn, es bestand eine klinische Kontraindikation.
  • Abraxane, in Kombination mit Gemcitabin, ist indiziert für die Erstlinienbehandlung von Patienten mit lokal fortgeschrittenem nicht resezierbarem oder metastasiertem Adenokarzinom des Pankreas.
  • Abraxane, in Kombination mit Carboplatin, ist indiziert für die Erstlinienbehandlung des nicht-kleinzelligen Lungenkarzinoms bei Patienten, bei denen keine potentiell kurative Operation und/oder Strahlentherapie möglich ist und die sich nicht für eine zielgerichtete molekulare Therapie eignen.

Paclitaxel

Pacritinib

According to the NCI website, pacritinib is an orally bioavailable inhibitor of Janus kinase 2 (JAK2) and the JAK2 mutant JAK2V617F with potential antineoplastic activity. Pacritinib competes with JAK2 for ATP binding, which may result in inhibition of JAK2 activation, inhibition of the JAK-STAT signaling pathway, and so caspase-dependent apoptosis. JAK2 is the most common mutated gene in bcr-abl-negative myeloproliferative disorders; the JAK2V617F gain-of-function mutation involves a valine-to-phenylalanine modification at position 617. Palbociclib – IBRANCE®

According to the NCI website, palbociclib is an orally available cyclin-dependent kinase (CDK) inhibitor with potential antineoplastic activity. Palbociclib selectively inhibits cyclin-dependent kinase 4 (CDK4) and 6 (CDK6), thereby inhibiting retinoblastoma (Rb) protein phosphorylation early in the G1 phase leading to cell cycle arrest. This suppresses DNA replication and decreases tumor cell proliferation. CDK4 and 6 are serine/threonine kinases that are upregulated in many tumor cell types and play a key role in the regulation of cell cycle progression. Indikationen/Anwendungsmöglichkeiten gemäss Compendium®

Ibrance ist angezeigt zur Behandlung von postmenopausalen Frauen mit Hormonrezeptor (HR)-positivem, humanen epidermalen Wachstumsfaktor-Rezeptor-2 (HER2)-negativem, lokal fortgeschrittenem oder metastasiertem Mammakarzinom:

  • in Kombination mit einem Aromatasehemmer;
  • in Kombination mit Fulvestrant bei Frauen, die zuvor eine endokrine Therapie erhielten.

Pamidronate/Aredia®

According to the NCI website pamidronate disodium is the disodium salt of the synthetic bisphosphonate pamidronate. Although its mechanism of action is not completely understood, pamidronate appears to adsorb to calcium phosphate crystals in bone, blocking their dissolution by inhibiting osteoclast-mediated bone resorption. This agent does not inhibit bone mineralization and formation.

Indikationen/Anwendungsmöglichkeiten gemäss Arzneimittel-Kompendium der Schweiz®:

Tumorinduzierte Hyperkalzämie.
Osteolytische Knochenmetastasen bei Mammakarzinom und Osteolyse bei multiplem Myelom.
Pamidronat verzögert das Fortschreiten der Osteolysen radiologisch gesehen, ohne dass relevante Komplikationen (wie z.B. Frakturen oder Mortalität) beeinflusst werden.

Bisphosphonat

Panitumumab - VECTIBIX®

According to the NCI website panitumumab is a human monoclonal antibody produced in transgenic mice that attaches to the transmembrane epidermal growth factor (EGF) receptor. Panitumumab may inhibit autocrine EGF stimulation of tumor cells that express the EGF receptor, thereby inhibiting tumor cell proliferation.

Indikationen/Anwendungsmöglichkeiten gemäss Compendium®:

Vectibix ist indiziert zur Behandlung von erwachsenen Patienten mit metastasiertem kolorektalem Karzinom (mCRC, metastatic colorectal cancer) mit RAS-Wildtyp

  • in der Erstlinientherapie in Kombination mit FOLFOX oder FOLFIRI.
  • in der Zweitlinientherapie in Kombination mit FOLFIRI bei Patienten, die in der Erstlinientherapie eine Fluoropyrimidin-haltige Chemotherapie erhalten haben (ausgenommen Irinotecan).

Monoclonal Antibodies

Panobinostat - FARYDAK®

Panobinostat - Farydak® ist ein Zimt-Hydroxamsäureanalog mit einer potentiellen antineoplastischen Aktivität. Panobinostat hemmt selektiv die Histon-Deacetylase (HDAC), induziert die Hyperacetylierung der Kern Histon-Proteine, was zur Modulation der Zellzyklusprotein-Expression führt und zum Zellzyklusstillstand in der G2/M Phase sowie zur Apoptose. Außerdem scheint dieses Mittel die Expression von Angiogenese-verwandten Genen zu modulieren, wie den durch Hypoxie induzierbaren Faktor-1alpha (HIF-1a) sowie den vaskulären endothelialen Wachstumsfaktor (VEGF), wodurch die Endothelzellen-Chemotaxis und die Invasion beeinträchtigt wird. HDAC ist ein Enzym, das Chromatin Histonproteine deacetyliert.

Indikationen/Anwendungsmöglichkeiten gemäss Compendium®:

  • Farydak ist in Kombination mit Bortezomib und Dexamethason für die Behandlung von Patienten mit multiplem Myelom indiziert, die mindestens zwei vorhergehende Therapien erhalten haben, inklusive Bortezomib und einen immunmodulatorischen Wirkstoff und nicht refraktär auf Bortezomib sind.

Parsaclisib

According to the NCI website, the PI3K-delta inhibitor INCB050465 (Parsaclisib) is an inhibitor of the delta isoform of phosphoinositide-3 kinase (PI3K) with potential antineoplastic activity. PI3K-delta inhibitor INCB050465 inhibits the delta isoform of PI3K and prevents the activation of the PI3K/AKT signaling pathway. This both decreases proliferation and induces cell death in PI3K-delta-overexpressing tumor cells. Unlike other isoforms of PI3K, PI3K-delta is expressed primarily in hematopoietic disease and cell lineages. The targeted inhibition of PI3K-delta is designed to preserve PI3K signaling in normal, non-neoplastic cells. Pazopanip - VOTRIENT®

Pazopanip - Votrient® ist das Hydrochloridsalz eines niedermolekularen Inhibitors von mehreren Protein-Tyrosin-Kinasen mit potentieller antineoplastischer Aktivität. Pazopanib hemmt selektiv die vaskulären endothelialen Wachstumsfaktor-Rezeptoren (VEGFR) -1, -2 und -3, c-kit und den Plättchenwachstumsfaktor-Rezeptor (PDGF-R). Dies kann in Tumoren, deren besagte Rezeptoren hochreguliert sind, zur Hemmung der Angiogenese führen.

Indikationen/Anwendungsmöglichkeiten gemäss Compendium®:

  • Behandlung des fortgeschrittenen und/oder metastasierenden Nierenzellkarzinoms.
  • Behandlung des fortgeschrittenen Weichteilsarkoms bei Progression nach Anthrazyklin-haltiger Chemotherapie oder anderen Kombinationen, wenn Anthrazyklin nicht in Frage kommt. Daten bei Liposarkom und GIST sind limitiert.

Pegfilgrastim/Neulasta®

According to the NCI website pegfilgrastim is a long-acting pegylated form of a recombinant therapeutic agent which is chemically identical to or similar to an endogenous human granulocyte colony-stimulating factor (G-CSF). Produced endogenously by monocytes, fibroblasts, and endothelial cells, G-CSF binds to and activates specific cell surface receptors, stimulating neutrophil progenitor proliferation and differentiation and selected neutrophil functions. Conjugation of the cytokine with a branched polyethylene glycol molecule (pegylation) significantly increases its therapeutic half-life.

Indikationen/Anwendungsmöglichkeiten gemäss Arzneimittel-Kompendium der Schweiz®:

Zur Verkürzung der Dauer von Neutropenien sowie zur Verminderung der Häufigkeit neutropenischen Fiebers bei Patienten, die wegen einer malignen Erkrankung mit zytotoxischer Chemotherapie behandelt werden (mit Ausnahme von chronischer myeloischer Leukämie und myelodysplastischem Syndrom).

Granulozyten-Kolonie stimulierende Faktor

Pembrolizumab - KEYTRUDA®

According to the NCI website pembrolizumab - Keytruda® is a humanized monoclonal IgG4 antibody directed against human cell surface receptor PD-1 (programmed death-1 or programmed cell death-1) with potential immunopotentiating activity. Upon administration, pembrolizumab binds to PD-1, an inhibitory signaling receptor expressed on the surface of activated T cells, and blocks the binding to and activation of PD-1 by its ligands, which results in the activation of T-cell-mediated immune responses against tumor cells.

Indikationen/Anwendungsmöglichkeiten gemäss Compendium®:

Melanom

  • Keytruda kann angewendet werden bei nicht resezierbarem oder metastasiertem Melanom bei Erwachsenen.
  • Keytruda ist als Monotherapie indiziert zur adjuvanten Behandlung von Erwachsenen mit vollständig reseziertem Melanom Stadium III (siehe «Klinische Wirksamkeit»).

Nicht-kleinzelliges Lungenkarzinom

  • Keytruda ist als Monotherapie indiziert zur Erstlinienbehandlung des metastasierten nicht-kleinzelligen Lungenkarzinoms (NSCLC) bei Erwachsenen, deren Tumore PD-L1 mit einem Tumour proportion score (TPS) ≥50% exprimieren und keine genomischen Tumoraberrationen vom EGFR oder ALK Typ haben.
  • Keytruda in Kombination mit Pemetrexed und Platin-Chemotherapie ist indiziert zur Erstlinienbehandlung des metastasierten nicht-plattenepithelialen NSCLC bei Erwachsenen, die keine genomischen Tumoraberrationen vom EGFR oder ALK Typ haben.
  • Keytruda in Kombination mit Carboplatin und entweder Paclitaxel oder Nab-Paclitaxel ist indiziert zur Erstlinienbehandlung des metastasierten plattenepithelialen NSCLC bei Erwachsenen.
  • Keytruda ist als Monotherapie indiziert zur Behandlung des fortgeschrittenen, metastasierten NSCLC nach vorangegangener Chemotherapie bei Erwachsenen, deren Tumore PD-L1 mit einem TPS ≥1% exprimieren. Patienten mit genomischen Tumoraberrationen vom EGFR oder ALK Typ sollten zudem eine für diese Aberrationen zugelassene Therapie erhalten haben, bevor sie mit Keytruda behandelt werden.

Kopf- und Halskarzinom

  • Keytruda ist in Kombination mit platin- und 5-fluorouracil-(5-FU)-haltiger Chemotherapie indiziert zur Behandlung von Erwachsenen mit rezidivierendem, nicht kurativ anzugehendem, lokal fortgeschrittenem oder metastasiertem PD-L1 exprimierendem Plattenepithelkarzinom des Kopf-Hals-Bereichs (HNSCC).
  • Keytruda ist als Monotherapie indiziert zur Behandlung von Erwachsenen mit rezidivierendem, nicht kurativ anzugehendem, lokal fortgeschrittenem oder metastasiertem HNSCC, die mit platin-basierter Chemotherapie vorbehandelt wurden und deren Tumore PD-L1 mit einem TPS ≥50% exprimieren.

Klassisches Hodgkin Lymphom

  • Keytruda ist indiziert zur Behandlung des refraktären oder rezidivierenden klassischen Hodgkin Lymphoms (rrcHL) bei Erwachsenen mit mindestens 3 Vorbehandlungen.

Primäres mediastinales grosszelliges B-Zell-Lymphom

Keytruda ist indiziert zur Behandlung des refraktären oder rezidivierenden primären mediastinalen grosszelligen B-Zell-Lymphoms (rrPMBCL) bei Erwachsenen

  • mit mindestens 2 Vorbehandlungen, von denen mindestens eine mit Rituximab durchgeführt wurde, und
  • die für eine autologe Stammzelltransplantation nicht in Frage kommen oder einen Rückfall nach Transplantation hatten.

Keytruda ist nicht indiziert zur Behandlung von Patienten mit PMBCL, die eine dringende zytoreduktive Therapie benötigen.

Urothelkarzinom

  • Keytruda ist indiziert zur Behandlung des lokal fortgeschrittenen oder metastasierten Urothelkarzinoms bei Erwachsenen, die mit platin-basierter Chemotherapie vorbehandelt wurden.

Tumore mit hoher Mikrosatelliteninstabilität

Keytruda als Monotherapie ist indiziert bei folgenden Tumoren mit hoher Mikrosatelliteninstabilität (MSI-H) oder fehlerhafter DNA-Mismatch-Reparatur (dMMR):

  • für die Behandlung von erwachsenen Patienten mit nicht reserzierbarem oder metastasiertem kolorektalem Karzinom (CRC) nach vorangegangener Fluoropyrimidin-basierter Therapie in Kombination mit Irinotecan oder Oxaliplatin;
  • für die Behandlung von erwachsenen Patienten mit metastasierten Endometriumkarzinomen, Magenkarzinomen, Dünndarmkarzinomen oder Gallengangskarzinomen, die nach Standardtherapie progredient sind und für die keine befriedigenden Alternativen in der Behandlung zur Verfügung stehen (siehe «Klinische Wirksamkeit»).

Nierenzellkarzinom (RCC)

  • Keytruda in Kombination mit Axitinib ist indiziert zur Erstlinienbehandlung des fortgeschrittenen Nierenzellkarzinoms (metastasiert oder rezidivierend) bei Erwachsenen (siehe «Klinische Wirksamkeit»).

Pemetrexed - ALIMTA®

According to the NCI website, pemetrexed is a drug used alone or with another drug to treat certain types of non-small cell lung cancer and malignant pleural mesothelioma. It is being studied in the treatment of other types of cancer. Pemetrexed disodium blocks DNA synthesis and may kill cancer cells. It is a type of folate antagonist. Also called Alimta and LY231514.

  • Monotherapie zur Erhaltungstherapie von Patienten mit lokal fortgeschrittenem oder metastasiertem nichtkleinzelligem Lungenkarzinom ausser bei überwiegender plattenepithelialer Histologie, direkt anschliessend an eine platinbasierte Chemotherapie von 4 Therapiezyklen bei fehlender Progression.
  • Monotherapie bei Patienten mit lokal fortgeschrittenem oder metastasiertem nichtkleinzelligem Lungenkarzinom ausser bei überwiegender plattenepithelialer Histologie nach vorangegangener Chemotherapie.

According to the NCI website, pemigatinib is an orally bioavailable inhibitor of the fibroblast growth factor receptor (FGFR) types 1, 2, and 3 (FGFR1/2/3), with potential antineoplastic activity. Pemigatinib binds to and inhibits FGFR1/2/3, which may result in the inhibition of FGFR1/2/3-related signal transduction pathways. This inhibits proliferation in FGFR1/2/3-overexpressing tumor cells. FGFR, a family of receptor tyrosine kinases upregulated in many tumor cell types, plays a key role in cellular proliferation, migration, and survival. Pentostatin

According to the NCI website pentostatin is a purine nucleotide analogue antibiotic isolated from the bacterium Streptomyces antibioticus. Also known as 2'-deoxycoformycin, pentostatin binds to and inhibits adenine deaminase (ADA), an enzyme essential to purine metabolism; ADA activity is greatest in cells of the lymphoid system with T-cells having higher activity than B-cells and T-cell malignancies higher ADA activity than B-cell malignancies. Pentostatin inhibition of ADA appears to result in elevated intracellular levels of dATP which may block DNA synthesis through the inhibition of ribonucleotide reductase. This agent may also inhibit RNA synthesis and may selectively deplete CD26+ lymphocytes.

Indikationen/Anwendungsmöglichkeiten gemäss Chemocare.com:

This medication is used in the treatment of hairy cell leukemia. It may also be used in certain non-hodgkin lymphomas, including cutaneous t-cell lymphoma.

Zytostatikum

Perifosine

According to the NCI website, perifosine is an orally active alkyl-phosphocholine compound with potential antineoplastic activity. Targeting cellular membranes, perifosine modulates membrane permeability, membrane lipid composition, phospholipid metabolism, and mitogenic signal transduction, resulting in cell differentiation and inhibition of cell growth. This agent also inhibits the anti-apoptotic mitogen-activated protein kinase (MAPK) pathway and modulates the balance between the MAPK and pro-apoptotic stress-activated protein kinase (SAPK/JNK) pathways, thereby inducing apoptosis. Perifosine has a lower gastrointestinal toxicity profile than the related agent miltefosine..

Pertuzumab - PERJETA®

According to the NCI website pertuzumab is a humanized recombinant monoclonal antibody directed against the extracellular dimerization domain of the HER-2 tyrosine kinase receptor. Binding of the antibody to the dimerization domain of the HER-2 tyrosine kinase receptor protein directly inhibits the ability of the HER-2 tyrosine kinase receptor protein (the most common pairing partner) to dimerize with other HER tyrosine kinase receptor proteins; inhibiting receptor protein dimerization prevents the activation of HER signaling pathways, resulting in tumor cell apoptosis.

Indikationen/Anwendungsmöglichkeiten gemäss Compendium®:

Metastasierter Brustkrebs

  • Perjeta ist in Kombination mit Herceptin und Docetaxel zur Behandlung von Patienten mit HER2-positivem metastasierendem oder lokal rezidivierendem nicht resezierbarem Brustkrebs indiziert, die noch keine Chemotherapie gegen ihre metastasierte Erkrankung erhalten haben.

Brustkrebs im Frühstadium

  • Perjeta ist in Kombination mit Herceptin und Docetaxel für die neoadjuvante Behandlung von Patienten mit HER2-positivem, lokal fortgeschrittenem, entzündlichem Brustkrebs oder Brustkrebs im Frühstadium mit hohem Rezidiv Risiko (entweder Tumorgrösse >2 cm Durchmesser oder mit Lymphknotenbefall) im Rahmen eines Therapieplanes für Brustkrebs im Frühstadium indiziert.
  • Perjeta ist in Kombination mit Herceptin und Chemotherapie für die adjuvante Behandlung von Patienten mit HER2-positivem Brustkrebs im Frühstadium mit hohem Rezidivrisiko indiziert (siehe Rubrik «Klinische Wirksamkeit»).

Pevonedistat

According to the NCI website, Pevonedistat is a small molecule inhibitor of Nedd8 activating enzyme (NAE) with potential antineoplastic activity. Pevonedistat binds to and inhibits NAE, which may result in the inhibition of tumor cell proliferation and survival. NAE activates Nedd8 (Neural precursor cell expressed, developmentally down-regulated 8), an ubiquitin-like (UBL) protein that modifies cellular targets in a pathway that is parallel to but distinct from the ubiquitin-proteasome pathway (UPP). Functioning in diverse regulatory activities, proteins conjugated to UBLs like Nedd8 typically are not targeted for proteasomal degradation. Check for active clinical trials using this agent. Polatuzumab vedotin - POLIVY® (USA, EU)

According to the NCI website Polatuzumab vedotin is an antibody-drug conjugate (ADC) composed of a monoclonal antibody directed against B-cell antigen receptor complex-associated protein beta chain (CD79B) conjugated, via a protease-cleavable peptide linker, to monomethyl auristatin E (MMAE), an auristatin derivative and a potent microtubule inhibitor, with potential antineoplastic activity. Upon administration, polatuzumab vedotin selectively binds to CD79B, a protein which is abundantly expressed on the surface of B-cells. Upon internalization and proteolytic cleavage, MMAE binds to tubulin and inhibits its polymerization, resulting in G2/M phase arrest and tumor cell apoptosis. CD79B, a component of the B-cell receptor (BCR), plays a key role in B-cell receptor signaling and is expressed on the surface of almost all types of malignant B-cells.

Indikation gemäss PDR:

  • Diffuse large B-cell lymphoma in adults whose cancer has relapsed or has not gotten better after at least two other therapies. It is used with bendamustine hydrochloride and rituximab.

Pomalidomid - IMNOVID®

Pomalidomid - Imnovid® ist oral bioverfügbares Derivat von Thalidomid mit potentiellen immunmodulierenden, antiangiogenen und antineoplastischen Aktivitäten. Obwohl der genaue Wirkmechanismus noch nicht vollständig aufgeklärt ist, scheint Pomalidomid die TNF-alpha-Produktion zu hemmen sowie die Aktivität von T-Zellen und natürliche Killerzellen (NK) und die Antikörper-abhängige zelluläre Zytotoxizität (ADCC) zu verbessern. Darüber hinaus kann Pomalidomid die Tumor-Angiogenese hemmen sowie den Zellzyklusstillstand in hierfür anfälligen Tumorzellpopulationen und die Erythropoese zu fördern.

Indikationen/Anwendungsmöglichkeiten gemäss Compendium®:

  • Imnovid in Kombination mit Bortezomib und Dexamethason ist indiziert zur Behandlung von erwachsenen Patienten mit multiplem Myelom (MM), welche mindestens eine vorgängige Therapie, inklusive Lenalidomid, erhielten.
  • Imnovid in Kombination mit Dexamethason ist indiziert zur Behandlung von rezidiviertem und refraktärem multiplem Myelom bei Patienten, welche mindestens zwei vorgängige Therapien erhielten (inklusive Lenalidomid und Bortezomib) und welche eine Progredienz zur letzten Therapie gezeigt haben.

Ponatinib - ICLUSIG®

Ponatinib - Iclusig® ist ein oral bioverfügbarer auf viele Ziele gerichteter Tyrosinkinase-Rezeptor (RTK)-Inhibitor mit potentiellen antiangiogenen und antineoplastischen Aktivitäten. Ponatinib Hydrochlorid hemmt sowohl die nicht-mutierten als auch alle mutierten Formen von Bcr-Abl, einschließlich T315I, der hoch therapieresistenten Missense-Mutation von Bcr-Abl. Dieses Mittel hemmt auch andere Tyrosinkinasen, einschließlich jener, die mit den vaskulären endothelialen Wachstumsfaktor-Rezeptoren (VEGFRs) und den Fibroblasten-Wachstumsfaktor-Rezeptoren (FGFRs) assoziiert sind; Darüber hinaus hemmt es den Tyrosinkinase-Rezeptor TIE2 und den FMS bezogenen Tyrosinkinase-Rezeptor-3 (Flt3). Die RTK Hemmung durch Ponatinib Hydrochlorid kann zur Hemmung der Zellproliferation und der Angiogenese führen und den Zelltod induzieren. Bcr-Abl ist eine vom Philadelphia-Chromosom kodierte Fusions Tyrosinkinase.

Iclusig ist indiziert bei erwachsenen Patienten mit

  • Philadelphia-Chromosom-positiver (Ph+) chronischer myeloischer Leukämie (in der chronischen Phase, akzelerierten Phase oder Blastenkrise), bei denen eine T315I-Mutation vorliegt, oder Ph+ akuter Lymphoblastenleukämie, bei denen eine T315I-Mutation vorliegt, oder
  • Ph+ chronischer myeloischer Leukämie (in der chronischen Phase, akzelerierten Phase oder Blastenkrise) oder Ph+ akuter Lymphoblastenleukämie, bei denen eine Behandlung mit anderen Inhibitoren der c-abl-Tyrosinkinase nicht geeignet ist.

According to the NCI website, Pralsetinib is an orally bioavailable selective inhibitor of mutant forms of and fusion products involving the proto-oncogene receptor tyrosine kinase RET, with potential antineoplastic activity. Procarbazin – NATULAN®

According to the NCI website, procarbazine hydrochloride, the hydrochloride salt of a methylhydrazine derivative has antineoplastic and mutagenic activities. Although the exact mode of cytotoxicity has not been elucidated, procarbazine, after metabolic activation, appears to inhibit the trans-methylation of methionine into transfer RNA (t-RNA), thereby preventing protein synthesis and consequently DNA and RNA synthesis. This agent may also undergo auto-oxidation, resulting in the formation of cytotoxic free radicals which damage DNA through an alkylation reaction. Check for active clinical trials using this agent. Prostvac

Prostvac - fowlpox-PSA-TRICOM vaccine is a cancer vaccine consisting of a recombinant fowlpox virus encoding human prostate-specific antigen (PSA) and TRICOM, a combination of three immunostimulants (i.e., B7.1, ICAM-1, and LFA-3). Administration of this agent may induce a cytotoxic T cell response against PSA-expressing tumor cells. Dendritic cells infected with TRICOM vectors greatly enhance naive T-cell activation and peptide-specific T-cell stimulation. Fowlpox virus is an attractive vector because its genome is easy to manipulate and it is replication incompetent in mammalian cells.
