According to the NCI website, encorafenib is an orally available Raf kinase inhibitor with potential antineoplastic activity. Encorafenib specifically inhibits Raf kinase, a serine/threonine enzyme in the RAF/mitogen-activated protein kinase kinase (MEK)/extracellular signal-related kinase (ERK) signaling pathway. By inhibiting the activation of the RAF/MEK/ERK signaling pathway, the administration of encorafenib may result in a decrease in proliferation of tumor cells. The Raf mutation BRAF V600E is frequently upregulated in a variety of human tumors and results in the constitutive activation of the RAF/MEK/ERK signaling pathway that regulates cellular proliferation and survival. According to the NCI website, Ensartinib is an orally available small molecule inhibitor of the receptor tyrosine kinase anaplastic lymphoma kinase (ALK) with potential antineoplastic activity. Upon oral administration, ensartinib binds to and inhibits ALK kinase, ALK fusion proteins and ALK point mutation variants. Inhibition of ALK leads to the disruption of ALK-mediated signaling and eventually inhibits tumor cell growth in ALK-expressing tumor cells. ALK belongs to the insulin receptor superfamily and plays an important role in nervous system development. ALK is not expressed in healthy adult human tissue but ALK dysregulation and gene rearrangements are associated with a series of tumors; ALK mutations are associated with acquired resistance to small molecule tyrosine kinase inhibitors. According to the NCI website, Entrectinib is an orally bioavailable inhibitor of the tyrosine kinases tropomyosin receptor kinases (Trk) A, B and C, C-ros oncogene 1 (ROS1) and anaplastic lymphoma kinase (ALK), with potential antineoplastic activity. Upon administration, entrectinib binds to and inhibits TrkA, TrkB, TrkC, ROS1 and ALK. Inhibition of these kinases may result in a disruption of TrkA-, TrkB-, TrkC-, ROS1-, and ALK-mediated signaling. This leads to an induction of apoptosis and an inhibition of tumor cell proliferation in tumor cells that express these kinases. - In Kombination mit LHRH-Agonisten zur Behandlung von Männern mit metastasiertem kastrationsresistentem Prostatakarzinom (CRPC) mit asymptomatischem oder mild symptomatischem Verlauf nach Versagen der Androgenentzugstherapie, und bei denen eine Chemotherapie noch nicht klinisch indiziert ist.
- In Kombination mit LHRH Agonisten zur Behandlung von Männern mit metastasierendem CRPC bei Progression unter oder nach Docetaxel Therapie.
Erlotinib-Hydrochlorid - Tarceva® ist das Hydrochloridsalz eines Chinazolin-Derivats und besitzt antineoplastische Eigenschaften. Im Wettbewerb mit Adenosintriphosphat bindet Erlotinib reversibel an die intrazelluläre katalytische Domäne der Tyrosinkinase des epidermalen Wachstumsfaktorrezeptors (EGFR). Dadurch wird die reversible EGFR-Phosphorylierung gehemmt und die mit der EGFR-Aktivierung assoziierte tumorfördernde Signaltransduktion blockiert.
Indikationen/Anwendungsmöglichkeiten gemäss Compendium®:
- Tarceva ist für die Behandlung von Patienten mit lokal fortgeschrittenem oder metastasierendem nicht-kleinzelligem Lungenkrebs indiziert, bei denen mindestens eine vorgängige Chemotherapie wirkungslos geblieben ist.
- Zur Erhaltungsbehandlung bei Patienten mit lokal fortgeschrittenem oder metastasiertem nicht-kleinzelligem Lungenkrebs im Anschluss an 4 Behandlungszyklen einer platinbasierten First-Linie Standardchemotherapie, wenn die Erkrankung nicht progredient ist und nicht ausreichend auf die Chemotherapie angesprochen hat.
- Tarceva ist für die Erstlinienbehandlung von Patienten mit lokal fortgeschrittenem oder metastasiertem nicht-kleinzelligem Lungenkarzinom (NSCLC) mit EGFR-aktivierenden Mutationen indiziert.

Everolimus - Afinitor® ist ein Derivat des natürlichen makrozyklischen Laktons Sirolimus mit immunsuppressiven und anti-angiogenen Eigenschaften. In Zellen bindet Everolimus an das Immunophilin FK-Bindungsprotein-12 (FKBP-12) und erzeugt damit einen immunsuppressiven Komplex, der die Aktivierung von mTOR (mammalian target of rapamycin) hemmt, eine Schlüssel-Kinase der Signalregulierung. Die Hemmung der mTOR Aktivierung führt zur Hemmung der T-Lymphozyten-Aktivierung und Proliferation, die wiederum mit der Antigen- und Zytokin-Stimulierung (IL-2, IL-4 und IL-15) sowie der Hemmung der Antikörperproduktion verbunden sind.
- In Kombination mit Exemestan zur Behandlung von postmenopausalen Frauen mit fortgeschrittenem, Hormonrezeptor positivem, HER 2 negativem Brustkrebs nach Versagen einer Behandlung mit Letrozol oder Anastrozol.
- Patienten mit fortgeschrittenen, progredienten, gut oder mässig differenzierten neuroendokrinen Tumoren pankreatischen Ursprungs.
- Patienten mit fortgeschrittenem Nierenzellkarzinom nach Versagen einer Behandlung mit Sunitinib oder Sorafenib.

According to the NCI website exemestane is a synthetic androgen analogue. Exemestane binds irreversibly to and inhibits the enzyme aromatase, thereby blocking the conversion of cholesterol to pregnenolone and the peripheral aromatization of androgenic precursors into estrogens. Indikationen/Anwendungsmöglichkeiten gemäss Compendium®:
- Adjuvante Behandlung von postmenopausalen Frauen mit invasivem frühem Mammakarzinom, welche Östrogen- oder Progesteronrezeptor-positiv oder von unbekanntem Rezeptorstatus sind, nach mindestens zwei Jahren initialer adjuvanter Tamoxifen-Therapie.
- Behandlung des fortgeschrittenen Mammakarzinoms bei Frauen mit natürlicher oder künstlich herbeigeführter Postmenopause nach Progression unter Antiöstrogenbehandlung.
- Aromasin ist auch angezeigt als Hormonbehandlung der dritten Wahl des fortgeschrittenen Mammakarzinoms bei Frauen mit natürlicher oder künstlich herbeigeführter Postmenopause nach Progression unter einer Behandlung mit Antiöstrogenen, nicht-steroidalen Aromatasehemmern oder Gestagenen.
