Bicalutamid - Casodex® sowie diverse Generika

Bicalutamid ist ein synthetisches nicht steroidales Antiandrogen. Es bindet sich kompetitiv an den cytosolischen Androgenrezeptor im Zielgewebe und hemmt dadurch die Bindung der Androgene an den Androgenrezeptor. Keine Verbindung findet statt mit den meisten mutierten Formen von Androgenrezeptoren.

Indikationen/Anwendungsmöglichkeiten gemäss Arzneimittelkompendium:

  • Casodex (50 mg/Tag) in Kombination mit LHRH-Analoga oder Orchiektomie beim fortgeschrittenen, metastasierenden Prostatakarzinom (M1). Die Erfahrungen in Kombination mit chirurgischer Kastration sind beschränkt.
  • Casodex (150 mg/Tag) beim lokal fortgeschrittenen Prostatakarzinom (T3–4, jedes N, M0; T1–2, N , M0) als Monotherapie, wenn die Standardtherapien radikale Prostatektomie, Radiotherapie, chirurgische oder medikamentöse Kastration nicht geeignet sind.
  • Casodex (150 mg/Tag) adjuvant nach radikaler Prostatektomie oder Strahlentherapie bei Patienten mit einem lokal fortgeschrittenen Prostatakarzinom und hohem Risiko einer Progression.
  • Beim metastasierenden Prostatakarzinom soll die Monotherapie mit Casodex 150 mg/Tag nicht angewendet werden

Bosutinib - Bosulif®

Bosutinib ist ein synthetisches Quinolon-Derivat und ein dualer inase-Hemmer der sowohl auf die  Abl- und auch die Src-Kinases abziehlt. Dies führt zu einer potentiell antineoplastischen Aktivität. Nicht wie Imatinib hemmt Bosutinib die Autophosphorylisierung der Abl- und Src-Kinasen.  Dies führt zu einer Hemmung des Zellwachstums sowie zur Apoptose. Aufgrund der dualen Wirkungsweise kann das Medikament eine Wirkung bei der resistenten CML entfalten sowie auch bei anderen myeloiden Erkrankungen sowie bei soliden Tumoren. 

Indikationen/Anwendungsmöglichkeiten gemäss Fachinformation des Arzneimittel-Kompendium der Schweiz®:

  • Bosulif ist indiziert für die Behandlung von erwachsenen Patienten mit neu diagnostizierter Philadelphia-Chromosom positiver chronischer myeloischer Leukämie (Ph+ CML) in der chronischen Phase.
  • Bosulif ist indiziert für die Behandlung von erwachsenen Patienten mit Philadelphia-Chromosom positiver chronisch myeloischer Leukämie (Ph+ CML) in der chronischen Phase, in der akzelerierten Phase sowie in der Blastenkrise, nach Vorbehandlung mit einem oder mehreren c-abl Tyrosinkinase Inhibitoren und wenn eine Behandlung mit Imatinib, Nilotinib oder Dasatinib nicht in Frage kommt.

Brigatinib - ALUNBRIG® (USA; EU)

 

According to the NCI website, brigatinib is an orally available inhibitor of receptor tyrosine kinases anaplastic lymphoma kinase (ALK) and the epidermal growth factor receptor (EGFR) with potential antineoplastic activity. Brigatinib binds to and inhibits ALK kinase and ALK fusion proteins as well as EGFR and mutant forms. This leads to the inhibition of ALK kinase and EGFR kinase, disrupts their signaling pathways and eventually inhibits tumor cell growth in susceptible tumor cells. In addition, brigatinib appears to overcome mutation-based resistance. ALK belongs to the insulin receptor superfamily and plays an important role in nervous system development; ALK dysregulation and gene rearrangements are associated with a series of tumors. Indikationen/Anwendungsmöglichkeiten gemäss EMEA

Alunbrig is a cancer medicine that is used to treat adults with a type of lung cancer called non-small cell lung cancer (NSCLC) who have been treated before with a cancer medicine called crizotinib.

Alunbrig is used on its own and only if the NSCLC is ‘ALK-positive’, which means that the cancer cells have certain changes affecting the gene that makes a protein called ALK (anaplastic lymphoma kinase).
